The ALSAfrica Network collection comprises genomic data from patients of African ancestry with amyotrophic lateral sclerosis (ALS) and related motor neuron disorders, clinically ascertained across four sites in South Africa, Kenya and Nigeria: the University of Cape Town, the University of the Witwatersrand, Aga Khan University and the University of Ibadan. This research effort investigates the genetic basis of ALS in African-ancestry populations, who remain substantially under-represented in global ALS genomics. The primary objective is twofold: to characterise the mutational spectrum of ALS in Africans, including single-nucleotide variants, insertions/deletions, structural variants and tandem-repeat expansions, thereby improving variant interpretation and ALS gene characterisation for these populations; and to harness the genetic diversity of African genomes to drive the discovery of novel ALS genes, variants and disease pathways. Data are generated by 30X PacBio HiFi long-read whole-genome sequencing. Individual-level data remain under the custodianship of the contributing site; the collection contributes aggregate, summary-level information (e.g. allele frequencies) to the...
